Physics explains the medical effect

Tirapazamine (TPZ) is a widely used antitumour drug whose detailed mechanism of action had remained unclear. Physicists at the University of Innsbruck — in a study involving this group — uncovered how low-energy electron attachment drives the decomposition of TPZ, revealing a "roaming mechanism" in which a hydroxyl radical forms as the dominant reaction product responsible for damaging tumour cell DNA. Quantum chemical calculations, combined with mass spectrometry experiments, gave a detailed picture of the reaction dynamics behind this widely used radio-chemotherapy agent.

The work was carried out jointly by the computational photophysics group of Milan Ončák and the inelastic electron scattering group of Stephan Denifl at the University of Innsbruck, with experimental work led by PhD student Eugene Arthur-Baidoo.
